{"id":2860,"date":"2020-12-07T12:27:10","date_gmt":"2020-12-07T06:57:10","guid":{"rendered":"https:\/\/www.classiciasacademy.com\/blog\/?p=2860"},"modified":"2020-12-07T12:27:49","modified_gmt":"2020-12-07T06:57:49","slug":"rt-lamp-for-covid-19-detection","status":"publish","type":"post","link":"https:\/\/www.classiciasacademy.com\/blog\/rt-lamp-for-covid-19-detection\/","title":{"rendered":"RT-LAMP- For Covid-19 Detection"},"content":{"rendered":"\n<h3 class=\"wp-block-heading\">In News<\/h3>\n\n\n\n<p>New technology for the detection of Covid-19 called <strong>RT-LAMP.<\/strong><\/p>\n\n\n\n<h3 class=\"wp-block-heading\">In-Detail<\/h3>\n\n\n\n<ul><li>As there is no vaccine\nfor Covid-19 infection there is a strong need for rapid, specific, and\nsensitive SARS-COV-2 detection technology to control and contain the virus.<\/li><li>RT-LAMP method is an\nideal method for diagnosis as it is simple, speed, and cost-effective.<\/li><\/ul>\n\n\n\n<h3 class=\"wp-block-heading\">Current Method<\/h3>\n\n\n\n<ul><li>The current method for\nSARS-COV-2 diagnosis is the <strong>RT-PCR (<\/strong><strong>real-time reverse\ntranscription-polymerase chain reaction.<\/strong><\/li><li>This test detects the\npresence of viral nucleic acids in nasopharyngeal swab samples.<\/li><li>But PCR test requires\ncomplex, costly equipment and requires extensive training for potential users.<\/li><li>The sensitivity and\nspecificity of the test is about 95% and there is a possibility of false-negative results.<\/li><li>The turnaround time is\nabout 10 hours and the result will be available only the next day.<\/li><li>In the remote areas,\nthe turnaround time may increase further depending on the distance the samples\nneed to travel.<\/li><li>To detect antigens,\nCOVID-19 serology tests are also used.<\/li><li>The serology tests are\neasy to use with rapid results and have minimal expenses.<\/li><li>The serology\nimmunoassay lacks the necessary accuracy due to its low sensitivity and high false-negative rates.<\/li><li>For one cycle, the\nRT-PCR test requires different temperatures.<\/li><li>The temperature of the\nsolution has to be changed from 92 degrees C to 56 degrees C and 72 degrees C\nfor every two minutes and this cycle has to be done repeatedly.<\/li><li>So, the PCR test\nrequires an expensive thermal cycler and also real-time PCR machines.<\/li><\/ul>\n\n\n\n<h3 class=\"wp-block-heading\">RT-LAMP<\/h3>\n\n\n\n<ul><li>RT-LAMP is a Reverse\nTranscriptase loop-mediated isothermal amplification technology.<\/li><li>This test is a\none-step nucleic acid amplification method.<\/li><li>This can multiply\nspecific sequences of RNA of the coronavirus.<\/li><li>By reverse\ntranscription, the RNA is first made into cDNA (copy DNA).<\/li><li>The DNA is then\namplified with the LAMP technology.<\/li><li>This technique is new\nto the Indian IVD industry but it has been used in western countries for the\npast five years.<\/li><li>In the RT-LAMP test,\nthe temperature required is 65 degrees C and the DNA amplification is done at\nconstant temperatures.<\/li><li>The quantity of DNA\namplified in the LAMP technology is a hundred thousand times more than in PCR.<\/li><li>The final assay is possible with a simple reaction and also removes the\nneed for costly real-time PCR machines.<\/li><li>The assay is so fast in the LAMP technology and can obtain the results\nwithin 30 minutes and the positive samples are amplified as early as 10\nminutes.<\/li><li>This test can report\nwith accuracy in substantially lower turnover time.<\/li><li>The reagents in the\nLAMP tests are to be stored at 4 degrees C.<\/li><li>The LAMP technology\ndoes not need laborious preparations and does not require complex expensive\nequipment.<\/li><li>LAMP test is cost-effective and the assay can be performed with minimum skill and\ninfrastructure. <\/li><li>LAMP technology has\nmany advantages when compared with other tests. <\/li><li>The Indian Council of\nMedical Research validated the LAMP technology with a sensitivity of 98.75% and\nspecificity of 100%.<\/li><li>\u25cf&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;\nThe LAMP-based kits have a positive predictive value of 100% and a\nnegative predictive value is 98.8% with an accuracy of 99.3%.<\/li><li>In India, Agappe\nDiagnostics developed the LAMP technology and their kit has been validated by\nthe ICMR for marketing and named it LUME Screen nCoV.<\/li><\/ul>\n\n\n\n<h3 class=\"wp-block-heading\">Conclusion<\/h3>\n\n\n\n<p>At present, a confirmatory, rapid, accurate, user-friendly,\nand low-cost molecular diagnostic method is a must to address the COVID-19\npandemic.
