{"id":2436,"date":"2019-12-11T10:54:43","date_gmt":"2019-12-11T05:24:43","guid":{"rendered":"https:\/\/www.classiciasacademy.com\/blog\/?p=2436"},"modified":"2019-12-11T10:54:43","modified_gmt":"2019-12-11T05:24:43","slug":"researchers-find-a-potent-molecule-to-fight-multidrug-resistant-bacteria","status":"publish","type":"post","link":"https:\/\/www.classiciasacademy.com\/blog\/researchers-find-a-potent-molecule-to-fight-multidrug-resistant-bacteria\/","title":{"rendered":"Researchers Find A Potent Molecule To Fight Multidrug-Resistant Bacteria"},"content":{"rendered":"\n<h3 class=\"wp-block-heading\">In\nNews <\/h3>\n\n\n\n<p>Researchers from the Indian Institute of Technology (IIT) Roorkee\nfound a new molecule that has the potential to kill multidrug-resistant\nBacteria. <\/p>\n\n\n\n<h3 class=\"wp-block-heading\">In-Detail\n<\/h3>\n\n\n\n<ul><li>For finding the new molecule,\nresearchers from IIT Roorkee have screened a small-molecule library of 11,000\ncompounds. <\/li><li>They found a potent molecule\nthat belongs to <strong>nitrofuran class of antibiotics &#8211; nitrofurantoin and\nfurazolidone &#8211; <\/strong>to exhibit broad-spectrum bacterial activity against\nmultidrug-resistant bacteria like E. Coli, Mycobacterium tuberculosis,\nKlebsiella pneumoniae, Acinetobacter baumannii, Staphylococcus aureus and Clostridium difficile. <\/li><\/ul>\n\n\n\n<h3 class=\"wp-block-heading\">The\nMolecule Testing<\/h3>\n\n\n\n<ul><li>The researchers have tested the\nmolecule in mice infected with sepsis-causing bacteria A. baumannii.<\/li><li>The molecule reduced the\nbacterial load significantly in the spleen, kidney, lungs and liver.<\/li><li>Only half the dose of the new\nmolecule compared to nitrofurantoin was enough to reduce the bacterial load. <\/li><li>The researchers found that the\nmolecule kills the bacteria in two ways. <\/li><li>It damages the DNA of the\nbacteria by inhibiting cell division. <\/li><li>When half the dose to kill the\nbacteria was used, the molecule hindered cell division in daughter cells\nleading to the bacteria forming into long filaments. <\/li><li>Because of the two pathways in\nwhich the molecule kills the bacteria, developing resistance by the bacteria\nwill take a very long time, explains the researchers. <\/li><li>They also found that, even at a\nlow 16-fold concentration, the molecule was able to kill E. Coli compared to\nnitrofurantoin. <\/li><li>The molecule was also effective\nagainst both gram-negative and gram-positive bacteria and anaerobic bacteria.<\/li><li>Also, persister bacteria that\nare dormant were also killed by the molecule. <\/li><\/ul>\n\n\n\n<h3 class=\"wp-block-heading\">Pro-Drug<\/h3>\n\n\n\n<ul><li>The researchers generated\nmutants of the molecule and developed a pro-drug. <\/li><li>This pro-drug will become\nactive only after getting inside the body of a bacteria, thereby limiting the\nscope of bacteria to develop resistance.<\/li><li>Also, the components formed by\nthe pro-drug are potent and short-lived, thus leaving less time for bacteria to\ndevelop resistance.<\/li><\/ul>\n\n\n\n<h3 class=\"wp-block-heading\">Doubly\nEffective<\/h3>\n\n\n\n<ul><li>The molecule was effective at\ninhibiting bacteria\u2019s ability to develop biofilm, the protective layer that\nbacteria use against antibiotics.<\/li><li>It is also effective at\ndisrupting already formed biofilms. <\/li><li>The researchers also found that\nthe bacteria\u2019s mechanism to flush out drugs from its body through efflux pumps\nwas ineffective against the new molecule.<\/li><li>Also, the tested the molecule\nto see whether reactive oxygen species (ROS) were generated to kill the\nbacteria. <\/li><li>They found that ROS was\nproduced after the bacterial kill.
